Medical Description

Vitamin D analog used in the treatment of vitamin D insufficiency/deficiency, prevention of osteoporosis and prevention of rickets in children.

Dosage & Administration

Adults: 200,000 IU IM single dose, may be repeated every 3–6 months as needed Children (rickets): Dose according to age and severity, under medical supervision Administered by deep intramuscular injection Oral vitamin D and calcium supplementation may be added if required

Side Effects

Usually well tolerated at recommended doses. Possible effects (especially with overdose): Hypercalcemia Hypercalciuria Nausea, vomiting Constipation Weakness, fatigue Polyuria, polydipsia

Contraindications Hypersensitivity to cholecalciferol Hypercalcemia Hypervitaminosis D Severe renal impairment with hypercalcemia Precautions Monitor serum calcium and vitamin D levels with repeated doses Use caution in renal impairment or sarcoidosis Avoid excessive intake of calcium or vitamin D from other sources

Drug Interactions

Thiazide diuretics → increased risk of hypercalcemia Anticonvulsants (phenytoin, carbamazepine) → reduced vitamin D effect Glucocorticoids → decreased vitamin D action Digitalis → increased risk of arrhythmias with hypercalcemia

Pregnancy & Lactation

Pregnancy: Use only if clearly needed and within recommended doses Lactation: Excreted in breast milk; generally safe at therapeutic doses
