Phosphodiesterase-5 enzyme inhibitor used in the treatment of erectile dysfunction, benign prostatic hyperplasia, pulmonary arterial hypertension and Raynaud's disease.

-May be administered with or without food. -When used on an as-needed basis, should be taken at least 30 minutes prior to sexual activity.

-Flushing, headache, nausea, myalgia, limb pain, respiratory tract infection, nasopharyngitis, hypertension, angina pectoris, chest pain, facial edema, hypotension, myocardial infarction, orthostatic hypotension, palpitations, peripheral edema, syncope, tachycardia, Dizziness, drowsiness, fatigue, hypoesthesia, insomnia, pain, paresthesia , vertigo, Diaphoresis, pruritus, skin rash, Increased gamma-glutamyl transferase, dyspepsia, gastroenteritis, gastroesophageal reflux disease, diarrhea, abdominal pain, dysphagia, esophagitis, gastritis, hemorrhoidal bleeding, loose stools, upper abdominal pain, vomiting, xerostomia, Urinary tract infection, prolonged erection, spontaneous erections, Rectal hemorrhage, abnormal hepatic function tests, Back pain, arthralgia, asthenia, neck pain, blurred vision, conjunctival hyperemia, conjunctivitis, eyelid edema, eye pain, increased lacrimation, vision color changes, Nasal congestion, paranasal sinus congestion, upper respiratory tract infection, cough, dyspnea, epistaxis and pharyngitis.

-If the creatinine clearance is 30 to 50 mL/minute: Initial: 2.5 mg once daily; maximum: 5 mg once daily. If the creatinine clearance is <30 mL/minute: Use not recommended. -Use is not recommended in case of severe hepatic impairment. -Use with caution in patients with bleeding disorders. -Use is not recommended in patients with hypotension (<90/50 mm Hg), uncontrolled hypertension (>170/100 mm Hg). -Use with caution in patients with active peptic ulcer disease. -Contraindicated in patients taking nitrates. When nitrate administration is medically necessary following the use of tadalafil, at least 48 hours should elapse after the tadalafil dose and before nitrate administration; close medical supervision is recommended. -Use tadalafil with caution in patients taking an alpha-blocking antihypertensive agent. -Contraindicated in patients suffering from hypersensitivity to tadalafil. -In case of over dose be ready to tell or show what was taken, how much and when it happened, and seek immediate medical attention. -Alcohol (Ethyl): May enhance the hypotensive effect of Phosphodiesterase 5 Inhibitors. -Alpha1-Blockers: Phosphodiesterase 5 Inhibitors may enhance the hypotensive effect of Alpha1-Blockers. -Blood pressure lowering agents: Phosphodiesterase 5 Inhibitors may enhance the hypotensive effect of blood pressure lowering agents. -Dapoxetine: May enhance the orthostatic hypotensive effect of Phosphodiesterase 5 Inhibitors. -Grapefruit juice may increase serum levels/toxicity of tadalafil.

-Tadalafil likely crosses the placenta. Information related to the use of tadalafil for pulmonary arterial hypertension in pregnant patients is limited. -It is not known if tadalafil is present in breast milk, the decision to breastfeed during therapy should consider the risk of infant exposure, the benefits of breastfeeding to the infant, and the benefits of treatment to the mother. -Ask your physician before taking any medication during pregnancy or lactation.
